Resumo

A caracterização dos alérgenos de peixes brasileiros ainda é deficiente. Este estudo foi conduzido com quatro indivíduos brasileiros que apresentaram reações imediatas após a ingestão de tilápia. A natureza mediada por IgE da hipersensibilidade foi demonstrada por testes cutâneos e por immunoblotting. Uma proteína ainda não caracterizada de 100 kDa foi identificada pelo immunoblotting IgE como o alérgeno do extrato natural não digerido da tilápia do Nilo (Oreochromis niloticus) geneticamente melhorada para aquacultura. Esta proteína foi susceptível à atividade péptica como demonstrado por SDS-PAGE, que mostrou uma banda única entre 12 e 19 kDa após a digestão péptica. Os testes cutâneos com o extrato digerido de tilápia não demonstraram nenhuma reação, assim como o immunoblotting IgE não demarcou nenhuma banda na corrida eletroforética do extrato digerido.

Abstract

Characterization of Brazilian fish allergens is still poor. This study was conducted with four Brazilian subjects who presented immediate reactions following tilapia ingestion. The IgE-mediated nature of their hypersensitivity was demonstrated by skin testing as well by IgE immunoblotting. A yet uncharacterized protein with approximately 100 kDa was identified by IgE immunoblotting as the allergen of the undigested natural extract of the genetically improved farmed Nile tilapia (Oreochromis niloticus) (GIFT). This protein was susceptible to pepsin activity, as demonstrated by SDS-PAGE, which showed a single protein band around 12 to 19 kDa after pepsin digestion. Skin testing with the digested GIFT extract did not demonstrate any reaction, and IgE immunoblotting did not stain any bands at the electrophoretic lane.
